Transport Media Overview
VTM (Viral Transport Medium)
VTM consists of a modified Hank’s-HEPES buffer solution enriched with bovine serum albumin, gelatin, antibiotics, and other stabilizers. A phenol red pH indicator is included to monitor medium integrity. The solution is osmotically balanced and buffered, ensuring the viability of viral specimens while preventing bacterial proliferation during transportation to the laboratory.
VTM-N (Inactivation Transport Medium)
VTM-N is a guanidine-based medium containing protein denaturants and nuclease inhibitors. It lyses cells, releasing and preserving nucleic acids (RNA & DNA) while inactivating viruses for safe handling. VTM-N maintains RNA and DNA stability at room temperature for up to 48 hours, but it is not suitable for viral culture or rapid antigen testing.
PBS and Normal Saline Solution
Phosphate Buffered Saline (PBS), including molecular-grade PBS or Delbecco’s PBS, can be used for sample collection and transport in molecular RT-PCR SARS-CoV-2 assays. If PBS is unavailable, normal saline may be used. The FDA recommends using a sterile glass or plastic vial containing 1–3 mL of PBS or normal saline. Specimens can be stored for up to 72 hours at 4°C.
